The role of affinity analysis in basic research
Affinity is a parameter that describes the strength of binding between ligand and analyte molecules. Essentially, the affinity between molecules is a non-covalent force, including attractive force, hydrogen bond force, hydrophobic force and so on. The affinity can be expressed by the affinity constant KD, and its calculation formula is as follows.
[Ab], [H] and [Ab•H] represent the corresponding molar concentrations of the two interacting substances. The affinity constant actually corresponds to the equilibrium constant of the chemical reaction. The higher the affinity constant of KD and the stronger the mutual binding force of the two substances to be tested.
In many basic researches, it is necessary to analyze the affinity of ligands and analytes, and provide a basis for the binding effect and strength between them. Generally, when the binding and dissociation rate between two substances is too fast to obtain an accurate binding and dissociation rate, only the way of affinity detection can be used under this condition.
